Human
specificity :
This assay has high sensitivity and excellent specificity for detection of ALPP. No significant cross-reactivity or interference between ALPP and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ALPP and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Quantitative Competitive. Sensitivity: 0.1 ng/mL.
products description :
Intended Uses: This ALPP ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ALPP. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ALPP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ALPP antibody and an ALPP-HRP conjugate. The assay sample and buffer are incubated together with ALPP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ALPP concentration since ALPP from samples and ALPP-HRP conjugate compete for the anti-ALPP antibody binding site. Since the number of sites is limited, as more sites are occupied by ALPP from the sample, fewer sites are left to bind ALPP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ALPP concentration in each sample is interpolated from this standard curve.

ncbi summary :
There are at least four distinct but related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney (tissue non-specific). The first three are located together on chromosome 2 while the tissue non-specific form is located on chromosome 1. The product of this gene is a membrane bound glycosylated enzyme, also referred to as the heat stable form, that is expressed primarily in the placenta although it is closely related to the intestinal form of the enzyme as well as to the placental-like form. The coding sequence for this form of alkaline phosphatase is unique in that the 3' untranslated region contains multiple copies of an Alu family repeat. In addition, this gene is polymorphic and three common alleles (type 1, type 2 and type 3) for this form of alkaline phosphatase have been well characterized. [provided by RefSeq, Jul 2008]
