specificity :
This assay has high sensitivity and excellent specificity for detection of Apo-C3. No significant cross-reactivity or interference between Apo-C3 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between Apo-C3 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 ng/mL.
other info2 :
Intended Uses: This Apo-C3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rabbit Apo-C3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: Apo-C3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Apo-C3 antibody and an Apo-C3-HRP conjugate. The assay sample and buffer are incubated together with Apo-C3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Apo-C3 concentration since Apo-C3 from samples and Apo-C3-HRP conjugate compete for the anti-Apo-C3 antibody binding site. Since the number of sites is limited, as more sites are occupied by Apo-C3 from the sample, fewer sites are left to bind Apo-C3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Apo-C3 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Apolipoprotein C-III is a very low density lipoprotein (VLDL) protein. APOC3 inhibits lipoprotein lipase and hepatic lipase; it is thought to delay catabolism of triglyceride-rich particles. The APOA1, APOC3 and APOA4 genes are closely linked in both rat and human genomes. The A-I and A-IV genes are transcribed from the same strand, while the A-1 and C-III genes are convergently transcribed. An increase in apoC-III levels induces the development of hypertriglyceridemia. [provided by RefSeq, Jul 2008]
uniprot summary :
APOC3: Inhibits lipoprotein lipase and hepatic lipase and decreases the uptake of lymph chylomicrons by hepatic cells. This suggests that it delays the catabolism of triglyceride-rich particles. Defects in APOC3 are the cause of hyperalphalipoproteinemia type 2 (HALP2). HALP2 is a condition characterized by high levels of high density lipoprotein (HDL) and increased HDL cholesterol levels. Belongs to the apolipoprotein C3 family. Protein type: Secreted; Secreted, signal peptide; Lipid-binding. Chromosomal Location of Human Ortholog: 11q23.3. Cellular Component: extracellular space; chylomicron; early endosome; extracellular region. Molecular Function: lipase inhibitor activity; cholesterol binding; phospholipid binding; enzyme regulator activity. Biological Process: response to drug; phototransduction, visible light; response to peptide hormone stimulus; cholesterol metabolic process; regulation of Cdc42 protein signal transduction; cholesterol efflux; lipoprotein metabolic process; G-protein coupled receptor protein signaling pathway; cholesterol homeostasis; triacylglycerol metabolic process; reverse cholesterol transport; phospholipid efflux; negative regulation of lipid catabolic process; lipoprotein transport; triacylglycerol catabolic process; negative regulation of lipoprotein lipase activity; negative regulation of receptor-mediated endocytosis; inflammatory response; negative regulation of lipid metabolic process; retinoid metabolic process; triacylglycerol mobilization; response to nutrient; negative regulation of fatty acid biosynthetic process. Disease: Apolipoprotein C-iii Deficiency
