Intended Uses: This ACCN3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ACCN3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ACCN3 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ACCN3 antibody and an ACCN3-HRP conjugate. The assay sample and buffer are incubated together with ACCN3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACCN3 concentration since ACCN3 from samples and ACCN3-HRP conjugate compete for the anti-ACCN3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACCN3 from the sample, fewer sites are left to bind ACCN3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACCN3 concentration in each sample is interpolated from this standard curve.

This gene encodes an enzyme involved in catalyzing the conversion of angiotensin I into a physiologically active peptide angiotensin II. Angiotensin II is a potent vasopressor and aldosterone-stimulating peptide that controls blood pressure and fluid-electrolyte balance. This enzyme plays a key role in the renin-angiotensin system. Many studies have associated the presence or absence of a 287 bp Alu repeat element in this gene with the levels of circulating enzyme or cardiovascular pathophysiologies. Multiple alternatively spliced transcript variants encoding different isoforms have been identified, and two most abundant spliced variants encode the somatic form and the testicular form, respectively, that are equally active.
