. Principle of the assay: GH2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GH2 antibody and an GH2-HRP conjugate. The assay sample and buffer are incubated together with GH2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GH2 concentration since GH2 from samples and GH2-HRP conjugate compete for the anti-GH2 antibody binding site. Since the number of sites is limited, as more sites are occupied by GH2 from the sample, fewer sites are left to bind GH2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GH2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. As in the case of its pituitary counterpart, growth hormone 1, the predominant isoform of this particular family member shows similar somatogenic activity, with reduced lactogenic activity.
