Intended Uses: This Abeta40 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat Abeta40. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: Abeta40 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-Abeta40 antibody and an Abeta40-HRP conjugate. The assay sample and buffer are incubated together with Abeta40-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Abeta40 concentration since Abeta40 from samples and Abeta40-HRP conjugate compete for the anti-Abeta40 antibody binding site. Since the number of sites is limited, as more sites are occupied by Abeta40 from the sample, fewer sites are left to bind Abeta40-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Abeta40 concentration in each sample is interpolated from this standard curve.

Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. In addition, two of the peptides are antimicrobial peptides, having been shown to have bacteriocidal and antifungal activities. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy).
