products description :
Intended Uses: This OPN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human OPN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: OPN ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for OPN. Standards or samples are then added to the microtiter plate wells and OPN if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of OPN present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for OPN are added to each well to "sandwich" the OPN immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain OPN and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The OPN concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]
uniprot summary :
osteopontin: Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Belongs to the osteopontin family. 5 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 4q22.1. Cellular Component: extracellular space; cell projection; apical part of cell; perinuclear region of cytoplasm; extracellular region. Molecular Function: protein binding; extracellular matrix binding; cytokine activity. Biological Process: osteoblast differentiation; neutrophil chemotaxis; extracellular matrix disassembly; extracellular matrix organization and biogenesis; response to vitamin D; response to steroid hormone stimulus; biomineral formation; negative regulation of collateral sprouting of intact axon in response to injury; positive regulation of bone resorption; decidualization; cell adhesion; inflammatory response; embryo implantation
