specificity :
This assay has high sensitivity and excellent specificity for detection of cTnT. No significant cross-reactivity or interference between cTnT and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between cTnT and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.

products description :
Intended Uses This cTnT ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse cTnT. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: cTnT ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-cTnT antibody and an cTnT-HRP conjugate. The assay sample and buffer are incubated together with cTnT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the cTnT concentration since cTnT from samples and cTnT-HRP conjugate compete for the anti-cTnT antibody binding site. Since the number of sites is limited, as more sites are occupied by cTnT from the sample, fewer sites are left to bind cTnT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The cTnT concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is the tropomyosin-binding subunit of the troponin complex, which is located on the thin filament of striated muscles and regulates muscle contraction in response to alterations in intracellular calcium ion concentration. Mutations in this gene have been associated with familial hypertrophic cardiomyopathy as well as with dilated cardiomyopathy. Transcripts for this gene undergo alternative splicing that results in many tissue-specific isoforms, however, the full-length nature of some of these variants has not yet been determined. uniprot summary :
TNNT2: Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity. Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart. Belongs to the troponin T family. 11 isoforms of the human protein are produced by alternative splicing. Protein type: Motility/polarity/chemotaxis; Motor. Chromosomal Location of Human Ortholog: 1q32. Cellular Component: sarcomere; troponin complex; cytosol; striated muscle thin filament. Molecular Function: troponin C binding; structural constituent of cytoskeleton; troponin I binding; ATPase activity; tropomyosin binding; actin binding. Biological Process: metabolic process; atrial cardiac muscle morphogenesis; positive regulation of ATPase activity; regulation of muscle contraction; ventricular cardiac muscle morphogenesis; sarcomere organization; response to calcium ion; negative regulation of ATPase activity; regulation of heart contraction; muscle filament sliding.
