products description :
Intended Uses: This SOCS3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse SOCS3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: SOCS3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SOCS3 antibody and an SOCS3-HRP conjugate. The assay sample and buffer are incubated together with SOCS3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SOCS3 concentration since SOCS3 from samples and SOCS3-HRP conjugate compete for the anti-SOCS3 antibody binding site. Since the number of sites is limited, as more sites are occupied by SOCS3 from the sample, fewer sites are left to bind SOCS3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SOCS3 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the STAT-induced STAT inhibitor (SSI), also known as suppressor of cytokine signaling (SOCS), family. SSI family members are cytokine-inducible negative regulators of cytokine signaling. The expression of this gene is induced by various cytokines, including IL6, IL10, and interferon (IFN)-gamma. The protein encoded by this gene can bind to JAK2 kinase, and inhibit the activity of JAK2 kinase. Studies of the mouse counterpart of this gene suggested the roles of this gene in the negative regulation of fetal liver hematopoiesis, and placental development. [provided by RefSeq, Jul 2008]
uniprot summary :
SOCS3: SOCS family proteins form part of a classical negative feedback system that regulates cytokine signal transduction. SOCS3 is involved in negative regulation of cytokines that signal through the JAK/STAT pathway. Inhibits cytokine signal transduction by binding to tyrosine kinase receptors including gp130, LIF, erythropoietin, insulin, IL12, GCSF and leptin receptors. Binding to JAK2 inhibits its kinase activity. Suppresses fetal liver erythropoiesis. Regulates onset and maintenance of allergic responses mediated by T-helper type 2 cells. Regulates IL-6 signaling in vivo. Probable substrate recognition component of a SCF-like ECS (Elongin BC- CUL2/5-SOCS-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. Seems to recognize IL6ST. Interacts with multiple activated proteins of the tyrosine kinase signaling pathway including IGF1 receptor, insulin receptor and JAK2. Binding to JAK2 is mediated through the KIR and SH2 domains to a phosphorylated tyrosine residue within the JAK2 JH1 domain. Binds specific activated tyrosine residues of the leptin, EPO, IL12, GSCF and gp130 receptors. Interaction with CSNK1E stabilizes SOCS3 protein. Component of the probable ECS(SOCS3) E3 ubiquitin-protein ligase complex which contains CUL5, RNF7/RBX2, Elongin BC complex and SOCS3. Interacts with CUL5, RNF7, TCEB1 and TCEB2. Interacts with CUL2. Interacts with FGFR3. Interacts with INSR. Widely expressed with high expression in heart, placenta, skeletal muscle, peripheral blood leukocytes, fetal and adult lung, and fetal liver and kidney. Lower levels in thymus. Protein type: Inhibitor. Chromosomal Location of Human Ortholog: 17q25.3. Cellular Component: cytoplasm; cytosol. Molecular Function: protein binding; protein kinase inhibitor activity. Biological Process: regulation of growth; cytokine and chemokine mediated signaling pathway; negative regulation of inflammatory response; negative regulation of protein kinase activity; negative regulation of insulin receptor signaling pathway; protein ubiquitination; positive regulation of cell differentiation; JAK-STAT cascade; negative regulation of JAK-STAT cascade; negative regulation of apoptosis.
