products description :
For samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate . INTENDED USE This C8b ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human C8b. This ELISA kit for research use only, not for therapeutic or diagnostic applications! . PRINCIPLE OF THE ASSAY C8b ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-C8b antibody and an C8b-HRP conjugate. The assay sample and buffer are incubated together with C8b-HRP conjugat e inpre-coated plate for one hour. After the incubation period, the wells a redecanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C8b concentration since C8b from samples and C8b-HRP conjugate compete for the anti-C8b antibody binding site. Since the number of sites is limited, as more sites are occupied by C8b from the sample, fewer sites are left to bind C8b-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C8b concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes one of the three subunits of the complement component 8 (C8) protein. C8 is composed of equimolar amounts of alpha, beta and gamma subunits, which are encoded by three separate genes. C8 is one component of the membrane attack complex, which mediates cell lysis, and it initiates membrane penetration of the complex. This protein mediates the interaction of C8 with the C5b-7 membrane attack complex precursor. In humans deficiency of this protein is associated with increased risk of meningococcal infections. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2013]
uniprot summary :
C8B: Constituent of the membrane attack complex (MAC) that plays a key role in the innate and adaptive immune response by forming pores in the plasma membrane of target cells. Defects in C8B are a cause of complement component 8 deficiency type 2 (C8D2). A rare defect of the complement classical pathway associated with susceptibility to severe recurrent infections, predominantly by Neisseria gonorrhoeae or Neisseria meningitidis. Belongs to the complement C6/C7/C8/C9 family. Protein type: Secreted, signal peptide; Secreted. Chromosomal Location of Human Ortholog: 1p32.2. Cellular Component: membrane attack complex; extracellular space; membrane; extracellular region; vesicle. Molecular Function: protein complex binding. Biological Process: cytolysis; complement activation, alternative pathway; regulation of complement activation; innate immune response; immune response; complement activation, classical pathway; complement activation.
