Principle of the Assay: NOX4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-NOX4 antibody and an NOX4 -HRP conjugate. The assay sample and buffer are incubated together with NOX4 -HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the NOX4 concentration since NOX4 from samples and NOX4 -HRP conjugate compete for the anti-NOX4 antibody binding site. Since the number of sites is limited, as more sites are occupied by NOX4 from the sample, fewer sites are left to bind NOX4 -HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The NOX4 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the NOX-family of enzymes that functions as the catalytic subunit the NADPH oxidase complex. The encoded protein is localized to non-phagocytic cells where it acts as an oxygen sensor and catalyzes the reduction of molecular oxygen to various reactive oxygen species (ROS). The ROS generated by this protein have been implicated in numerous biological functions including signal transduction, cell differentiation and tumor cell growth. A pseudogene has been identified on the other arm of chromosome 11. Alternative splicing results in multiple transcript variants.[provided by RefSeq, Jan 2009]
