Principle of the Assay: PRF1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PRF1 antibody and an PRF1-HRP conjugate. The assay sample and buffer are incubated together with PRF1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PRF1 concentration since PRF1 from samples and PRF1-HRP conjugate compete for the anti-PRF1 antibody binding site. Since the number of sites is limited, as more sites are occupied by PRF1 from the sample, fewer sites are left to bind PRF1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PRF1 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene has structural and functional similarities to complement component 9 (C9). Like C9, this protein creates transmembrane tubules and is capable of lysing non-specifically a variety of target cells. This protein is one of the main cytolytic proteins of cytolytic granules, and it is known to be a key effector molecule for T-cell- and natural killer-cell-mediated cytolysis. Defects in this gene cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare and lethal autosomal recessive disorder of early childhood. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq, Jul 2008]
