Intended Uses: This STAT6 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse STAT6. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay STAT6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-STAT6 antibody and an STAT6-HRP conjugate. The assay sample and buffer are incubated together with STAT6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the STAT6 concentration since STAT6 from samples and STAT6-HRP conjugate compete for the anti-STAT6 antibody binding site. Since the number of sites is limited, as more sites are occupied by STAT6 from the sample, fewer sites are left to bind STAT6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The STAT6 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the STAT family of transcription factors. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein plays a central role in exerting IL4 mediated biological responses. It is found to induce the expression of BCL2L1/BCL-X(L), which is responsible for the anti-apoptotic activity of IL4. Knockout studies in mice suggested the roles of this gene in differentiation of T helper 2 (Th2) cells, expression of cell surface markers, and class switch of immunoglobulins.
