Intended Uses: This sVCAM-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine sVCAM-1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: sVCAM-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-sVCAM-1 antibody and an sVCAM-1-HRP conjugate. The assay sample and buffer are incubated together with sVCAM-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the sVCAM-1 concentration since sVCAM-1 from samples and sVCAM-1-HRP conjugate compete for the anti-sVCAM-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by sVCAM-1 from the sample, fewer sites are left to bind sVCAM-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The sVCAM-1 concentration in each sample is interpolated from this standard curve.

Many different human endogenous retrovirus (HERV) families are expressed in normal placental tissue at high levels, suggesting that HERVs are functionally important in reproduction. This gene is part of an HERV provirus on chromosome 7 that has inactivating mutations in the gag and pol genes. This gene is the envelope glycoprotein gene which appears to have been selectively preserved. The gene's protein product is expressed in the placental syncytiotrophoblast and is involved in fusion of the cytotrophoblast cells to form the syncytial layer of the placenta. The protein has the characteristics of a typical retroviral envelope protein, including a furin cleavage site that separates the surface (SU) and transmembrane (TM) proteins which form a heterodimer.
