specificity :
This assay has high sensitivity and excellent specificity for detection of ADAMT1M4. No significant cross-reactivity or interference between ADAMT1M4 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ADAMT1M4 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 pg/mL.
other info2 :
Intended Uses: This ADAMT1M4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ADAMT1M4. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: ADAMT1M4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADAMT1M4 antibody and an ADAMT1M4-HRP conjugate. The assay sample and buffer are incubated together with ADAMT1M4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADAMT1M4 concentration since ADAMT1M4 from samples and ADAMT1M4-HRP conjugate compete for the anti-ADAMT1M4 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADAMT1M4 from the sample, fewer sites are left to bind ADAMT1M4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAMT1M4 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme encoded by this gene lacks a C-terminal TS motif. It is responsible for the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracellular matrix protein. The cleavage of aggrecan and brevican suggests key roles of this enzyme in arthritic disease and in the central nervous system, potentially, in the progression of glioma. uniprot summary :
ADAMTS4: Cleaves aggrecan, a cartilage proteoglycan, and may be involved in its turnover. May play an important role in the destruction of aggrecan in arthritic diseases. Could also be a critical factor in the exacerbation of neurodegeneration in Alzheimer disease. Cleaves aggrecan at the 392-Glu- -Ala-393 site.
