INTENDED USE This PDGF-D ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse PDGF-D. This ELISA kit for research use only, not for therapeutic or diagnostic applications! . PRINCIPLE OF THE ASSAY PDGF-D ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PDGF-D antibody and an PDGF-D-HRP conjugate. The assay sample and buffer are incubated together with PDGF-D-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PDGF-D concentration since PDGF-D from samples and PDGF-D-HRP conjugate compete for the anti-PDGF-D antibody binding site. Since the number of sites is limited, as more sites are occupied by PDGF-D from the sample, fewer sites are left to bind PDGF-D-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PDGF-D concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the platelet-derived growth factor family. The four members of this family are mitogenic factors for cells of mesenchymal origin and are characterized by a core motif of eight cysteines, seven of which are found in this factor. This gene product only forms homodimers and, therefore, does not dimerize with the other three family members. It differs from alpha and beta members of this family in having an unusual N-terminal domain, the CUB domain. Two splice variants have been identified for this gene. [provided by RefSeq, Jul 2008]
