Principle of the assay: ABCA7 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ABCA7 antibody and an ABCA7-HRP conjugate. The assay sample and buffer are incubated together with ABCA7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ABCA7 concentration since ABCA7 from samples and ABCA7-HRP conjugate compete for the anti-ABCA7 antibody binding site. Since the number of sites is limited, as more sites are occupied by ABCA7 from the sample, fewer sites are left to bind ABCA7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ABCA7 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the ABC1 subfamily. Members of the ABC1 subfamily comprise the only major ABC subfamily found exclusively in multicellular eukaryotes. This full transporter has been detected predominantly in myelo-lymphatic tissues with the highest expression in peripheral leukocytes, thymus, spleen, and bone marrow.
