Intended Uses: This FBN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine FBN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: FBN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FBN antibody and an FBN-HRP conjugate. The assay sample and buffer are incubated together with FBN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBN concentration since FBN from samples and FBN-HRP conjugate compete for the anti-FBN antibody binding site. Since the number of sites is limited, as more sites are occupied by FBN from the sample, fewer sites are left to bind FBN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBN concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of the ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbxs class and contains an F-box domain. This protein is highly expressed during muscle atrophy, whereas mice deficient in this gene were found to be resistant to atrophy. This protein is thus a potential drug target for the treatment of muscle atrophy. Alternative splicing results in multiple transcript variants encoding different isoforms.
