Principle of the assay: CLEC4E ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLEC4E antibody and an CLEC4E-HRP conjugate. The assay sample and buffer are incubated together with CLEC4E-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLEC4E concentration since CLEC4E from samples and CLEC4E-HRP conjugate compete for the anti-CLEC4E antibody binding site. Since the number of sites is limited, as more sites are occupied by CLEC4E from the sample, fewer sites are left to bind CLEC4E-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLEC4E concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signalling, glycoprotein turnover, and roles in inflammation and immune response. The encoded type II transmembrane protein is a downstream target of CCAAT/enhancer binding protein (C/EBP), beta (CEBPB) and may play a role in inflammation. Alternative splice variants have been described but their full-length sequence has not been determined. This gene is closely linked to other CTL/CTLD superfamily members on chromosome 12p13 in the natural killer gene complex region. [provided by RefSeq, Jul 2008]
