specificity :
This assay has high sensitivity and excellent specificity for detection of CLEC4E. No significant cross-reactivity or interference between CLEC4E and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CLEC4E and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 pg/mL.
other info2 :
Intended Uses: This CLEC4E ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse CLEC4E. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: CLEC4E ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLEC4E antibody and an CLEC4E-HRP conjugate. The assay sample and buffer are incubated together with CLEC4E-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLEC4E concentration since CLEC4E from samples and CLEC4E-HRP conjugate compete for the anti-CLEC4E antibody binding site. Since the number of sites is limited, as more sites are occupied by CLEC4E from the sample, fewer sites are left to bind CLEC4E-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLEC4E concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily. Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signalling, glycoprotein turnover, and roles in inflammation and immune response. The encoded type II transmembrane protein is a downstream target of CCAAT/enhancer binding protein (C/EBP), beta (CEBPB) and may play a role in inflammation. Alternative splice variants have been described but their full-length sequence has not been determined. This gene is closely linked to other CTL/CTLD superfamily members on chromosome 12p13 in the natural killer gene complex region. [provided by RefSeq, Jul 2008]
uniprot summary :
CLEC4E: C-type lectin that functions as cell-surface receptor for a wide variety of ligands such as damaged cells, fungi and mycobacteria. Plays a role in the recognition of pathogenic fungi, such as Candida albicans. The detection of mycobacteria is via trehalose 6,6 -dimycolate (TDM), a cell wall glycolipid. Specifically recognizes alpha-mannose residues on pathogenic fungi of the genus Malassezia. Recognizes also SAP130, a nuclear protein, that is released by dead or dying cells. Transduces signals through an ITAM-containing adapter protein, Fc receptor gamma chain /FCER1G. Induces secretion of inflammatory cytokines through a pathway that depends on SYK, CARD9 and NF-kappa-B. Protein type: Membrane protein, integral; Receptor, misc. Chromosomal Location of Human Ortholog: 12p13.31. Cellular Component: integral to membrane. Molecular Function: receptor activity; carbohydrate binding. Biological Process: immune response; positive regulation of cytokine secretion
