Intended Uses: This AbetaA4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey AbetaA4. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: AbetaA4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AbetaA4 antibody and an AbetaA4-HRP conjugate. The assay sample and buffer are incubated together with AbetaA4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AbetaA4 concentration since AbetaA4 from samples and AbetaA4-HRP conjugate compete for the anti-AbetaA4 antibody binding site. Since the number of sites is limited, as more sites are occupied by AbetaA4 from the sample, fewer sites are left to bind AbetaA4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AbetaA4 concentration in each sample is interpolated from this standard curve.

This gene encodes a cell surface receptor and transmembrane precursor protein that is cleaved by secretases to form a number of peptides. Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. In addition, two of the peptides are antimicrobial peptides, having been shown to have bacteriocidal and antifungal activities. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy).
