products description :
For samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate . INTENDED USE This AQP-5 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rabbit AQP-5. This ELISA kit for research use only, not for therapeutic or diagnostic applications! . PRINCIPLE OF THE ASSAY AQP-5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AQP-5 antibody and an AQP-5-HRP conjugate. The assay sample and buffer are incubated together with AQP-5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AQP-5 concentration since AQP-5 from samples and AQP-5-HRP conjugate compete for the anti-AQP-5 antibody binding site. Since the number of sites is limited, as more sites are occupied by AQP-5 from the sample, fewer sites are left to bind AQP-5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AQP-5 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Aquaporin 5 (AQP5) is a water channel protein. Aquaporins are a family of small integral membrane proteins related to the major intrinsic protein (MIP or AQP0). Aquaporin 5 plays a role in the generation of saliva, tears and pulmonary secretions. AQP0, AQP2, AQP5, and AQP6 are closely related and all map to 12q13. [provided by RefSeq, Jul 2008]
