reactivity :
Human
specificity :
This assay has high sensitivity and excellent specificity for detection of CHI3L1. No significant cross-reactivity or interference between CHI3L1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CHI3L1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
other info2 :
Intended Uses: This CHI3L1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CHI3L1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: CHI3L1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CHI3L1 antibody and an CHI3L1-HRP conjugate. The assay sample and buffer are incubated together with CHI3L1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CHI3L1 concentration since CHI3L1 from samples and CHI3L1-HRP conjugate compete for the anti-CHI3L1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CHI3L1 from the sample, fewer sites are left to bind CHI3L1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CHI3L1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Chitinases catalyze the hydrolysis of chitin, which is an abundant glycopolymer found in insect exoskeletons and fungal cell walls. The glycoside hydrolase 18 family of chitinases includes eight human family members. This gene encodes a glycoprotein member of the glycosyl hydrolase 18 family. The protein lacks chitinase activity and is secreted by activated macrophages, chondrocytes, neutrophils and synovial cells. The protein is thought to play a role in the process of inflammation and tissue remodeling. [provided by RefSeq, Sep 2009]
uniprot summary :
CHI3L1: Carbohydrate-binding lectin with a preference for chitin. May play a role in defense against pathogens, or in tissue remodeling. May play an important role in the capacity of cells to respond to and cope with changes in their environment. A genetic variation in CHI3L1 is associated with susceptibility to asthma-related traits type 7 (ASRT7). Asthma-related traits include clinical symptoms of asthma, such as coughing, wheezing and dyspnea, bronchial hyperresponsiveness (BHR) as assessed by methacholine challenge test, serum IgE levels, atopy, and atopic dermatitis. Belongs to the glycosyl hydrolase 18 family. Protein type: Secreted, signal peptide; Cell adhesion; Secreted. Chromosomal Location of Human Ortholog: 1q32.1. Cellular Component: proteinaceous extracellular matrix; extracellular space; endoplasmic reticulum; perinuclear region of cytoplasm; cytoplasm. Molecular Function: extracellular matrix structural constituent; chitin binding; chitinase activity; carbohydrate binding. Biological Process: positive regulation of protein kinase B signaling cascade; positive regulation of angiogenesis; apoptosis; response to mechanical stimulus; cartilage development; carbohydrate metabolic process; activation of NF-kappaB-inducing kinase; immune response; chitin catabolic process; inflammatory response; lung development. Disease: Schizophrenia; Asthma-related Traits, Susceptibility To, 7
