products description :
For samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate . INTENDED USE This A? 42 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse A? 42. This ELISA kit for research use only, not for therapeutic or diagnostic applications! . PRINCIPLE OF THE ASSAY A? 42 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-A? 42 antibody and an A? 42-HRP conjugate. The assay sample and buffer are incubated together with A? 42-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the A? 42 concentration since A? 42 from samples and A? 42-HRP conjugate compete for the anti-A? 42 antibody binding site. Since the number of sites is limited, as more sites are occupied by A? 42 from the sample, fewer sites are left to bind A? 42-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The A? 42 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a cell surface receptor and transmembrane precursor protein that is cleaved by secretases to form a number of peptides. Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. In addition, two of the peptides are antimicrobial peptides, having been shown to have bacteriocidal and antifungal activities. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy). Multiple transcript variants encoding several different isoforms have been found for this gene. [provided by RefSeq, Aug 2014]
