Human
specificity :
This assay has high sensitivity and excellent specificity for detection of BMP-10. No significant cross-reactivity or interference between BMP-10 and analogues was observed. NOTE: cross-reactivity detection between BMP-10 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This BMP-10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human BMP-10. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay BMP-10 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for BMP-10. Standards or samples are then added to the microtiter plate wells and BMP-10 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of BMP-10 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for BMP-10 are added to each well to "sandwich" the BMP-10 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain BMP-10 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BMP-10 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is a member of the TGF-beta family of growth factors. Data suggest that the similar protein in mouse plays an important role in trabeculation of the embryonic heart. In human, this protein may signal through receptor serine/threonine kinases. [provided by RefSeq, Jul 2008]
uniprot summary :
BMP10: Required for maintaining the proliferative activity of embryonic cardiomyocytes by preventing premature activation of the negative cell cycle regulator CDKN1C/p57KIP and maintaining the required expression levels of cardiogenic factors such as MEF2C and NKX2-5. Acts as a ligand for ACVRL1/ALK1, BMPR1A/ALK3 and BMPR1B/ALK6, leading to activation of SMAD1, SMAD5 and SMAD8 transcription factors. Inhibits endothelial cell migration and growth. Belongs to the TGF-beta family. Protein type: Secreted; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 2p13.3. Cellular Component: extracellular space; cell surface; cytoplasm; extracellular region; Z disc. Molecular Function: receptor serine/threonine kinase binding; protein binding; telethonin binding; growth factor activity; hormone activity; cytokine activity; transforming growth factor beta receptor binding. Biological Process: adult heart development; extracellular matrix organization and biogenesis; atrial cardiac muscle morphogenesis; positive regulation of transcription, DNA-dependent; ventricular cardiac muscle cell development; activin receptor signaling pathway; cardiac muscle cell proliferation; sarcomere organization; regulation of apoptosis; BMP signaling pathway; regulation of MAPKKK cascade; ventricular cardiac muscle morphogenesis; negative regulation of cell growth; cell adhesion; negative regulation of cell migration; positive regulation of cardiac muscle cell proliferation
