reactivity :
Human
specificity :
This assay has high sensitivity and excellent specificity for detection of TDP-43. No significant cross-reactivity or interference between TDP-43 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TDP-43 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Sandwich. Sensitivity: 0.1 ng/mL
products description :
Intended Uses: This TDP-43 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human TDP-43. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: TDP-43 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for TDP-43. Standards or samples are then added to the microtiter plate wells and TDP-43 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of TDP-43 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for TDP-43 are added to each well to "sandwich" the TDP-43 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain TDP-43 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TDP-43 concentration in each sample is interpolated from this standard curve.

uniprot summary :
TARDBP: DNA and RNA-binding protein which regulates transcription and splicing. Involved in the regulation of CFTR splicing. It promotes CFTR exon 9 skipping by binding to the UG repeated motifs in the polymorphic region near the 3 -splice site of this exon. The resulting aberrant splicing is associated with pathological features typical of cystic fibrosis. May also be involved in microRNA biogenesis, apoptosis and cell division. Can repress HIV-1 transcription by binding to the HIV-1 long terminal repeat. Stabilizes the low molecular weight neurofilament (NFL) mRNA through a direct interaction with the 3 UTR. Homodimer. Binds specifically to pyrimidine-rich motifs of TAR DNA and to single stranded TG repeated sequences. Binds to RNA, specifically to UG repeated sequences with a minimun of six contiguous repeats. Interacts with ATNX2; the interaction is RNA-dependent. Ubiquitously expressed. In particular, expression is high in pancreas, placenta, lung, genital tract and spleen. 3 isoforms of the human protein are produced by alternative splicing. Protein type: RNA-binding. Chromosomal Location of Human Ortholog: 1p36.22. Cellular Component: nucleoplasm; perichromatin fibrils; cytoplasm; nuclear speck; nucleus. Molecular Function: identical protein binding; protein binding; mRNA 3 -UTR binding; RNA binding; double-stranded DNA binding; nucleotide binding; transcription factor activity. Biological Process: transcription from RNA polymerase II promoter; RNA splicing; positive regulation of insulin secretion; positive regulation of transcription from RNA polymerase II promoter; mRNA processing. Disease: Amyotrophic Lateral Sclerosis 10, With Or Without Frontotemporal Dementia
