Intended Uses: This BAG2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse BAG2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: BAG2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BAG2 antibody and an BAG2-HRP conjugate. The assay sample and buffer are incubated together with BAG2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BAG2 concentration since BAG2 from samples and BAG2-HRP conjugate compete for the anti-BAG2 antibody binding site. Since the number of sites is limited, as more sites are occupied by BAG2 from the sample, fewer sites are left to bind BAG2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BAG2 concentration in each sample is interpolated from this standard curve.

BAG proteins compete with Hip for binding to the Hsc70/Hsp70 ATPase domain and promote substrate release. All the BAG proteins have an approximately 45-amino acid BAG domain near the C terminus but differ markedly in their N-terminal regions. The predicted BAG2 protein contains 211 amino acids. The BAG domains of BAG1, BAG2, and BAG3 interact specifically with the Hsc70 ATPase domain in vitro and in mammalian cells. All 3 proteins bind with high affinity to the ATPase domain of Hsc70 and inhibit its chaperone activity in a Hip-repressible manner. [provided by RefSeq, Jul 2008]
