specificity :
This assay has high sensitivity and excellent specificity for detection of KLKB1. No significant cross-reactivity or interference between KLKB1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between KLKB1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This KLKB1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human KLKB1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: KLKB1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-KLKB1 antibody and an KLKB1-HRP conjugate. The assay sample and buffer are incubated together with KLKB1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KLKB1 concentration since KLKB1 from samples and KLKB1-HRP conjugate compete for the anti-KLKB1 antibody binding site. Since the number of sites is limited, as more sites are occupied by KLKB1 from the sample, fewer sites are left to bind KLKB1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KLKB1 concentration in each sample is interpolated from this standard curve.

uniprot summary :
KLKB1: The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency); also known as Fletcher factor deficiency. This disorder is a blood coagulation defect. Belongs to the peptidase S1 family. Plasma kallikrein subfamily. Protein type: Secreted, signal peptide; Secreted; EC 3.4.21.34; Protease. Cellular Component: extracellular space; extracellular region. Molecular Function: peptidase activity; hydrolase activity; serine-type peptidase activity; serine-type endopeptidase activity; catalytic activity. Biological Process: fibrinolysis; hemostasis; positive regulation of fibrinolysis; inflammatory response; proteolysis; blood coagulation; plasminogen activation
