Principle of the assay: CSTD ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CSTD antibody and an CSTD-HRP conjugate. The assay sample and buffer are incubated together with CSTD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CSTD concentration since CSTD from samples and CSTD-HRP conjugate compete for the anti-CSTD antibody binding site. Since the number of sites is limited, as more sites are occupied by CSTD from the sample, fewer sites are left to bind CSTD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CSTD concentration in each sample is interpolated from this standard curve.

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a protein found in saliva and tears.
