Principle of the assay: BCL11A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BCL11A antibody and an BCL11A-HRP conjugate. The assay sample and buffer are incubated together with BCL11A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BCL11A concentration since BCL11A from samples and BCL11A-HRP conjugate compete for the anti-BCL11A antibody binding site. Since the number of sites is limited, as more sites are occupied by BCL11A from the sample, fewer sites are left to bind BCL11A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BCL11A concentration in each sample is interpolated from this standard curve.

This gene encodes a C2H2 type zinc-finger protein by its similarity to the mouse Bcl11a/Evi9 protein. The corresponding mouse gene is a common site of retroviral integration in myeloid leukemia, and may function as a leukemia disease gene, in part, through its interaction with BCL6. During hematopoietic cell differentiation, this gene is down-regulated. It is possibly involved in lymphoma pathogenesis since translocations associated with B-cell malignancies also deregulates its expression. Multiple transcript variants encoding several different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
