products description :
Intended Uses: This ATF-3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ATF-3. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ATF-3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ATF-3 antibody and an ATF-3-HRP conjugate. The assay sample and buffer are incubated together with ATF-3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATF-3 concentration since ATF-3 from samples and ATF-3-HRP conjugate compete for the anti-ATF-3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATF-3 from the sample, fewer sites are left to bind ATF-3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATF-3 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the mammalian activation transcription factor/cAMP responsive element-binding (CREB) protein family of transcription factors. This gene is induced by a variety of signals, including many of those encountered by cancer cells, and is involved in the complex process of cellular stress response. Multiple transcript variants encoding different isoforms have been found for this gene. It is possible that alternative splicing of this gene may be physiologically important in the regulation of target genes. [provided by RefSeq, Apr 2011]
uniprot summary :
ATF-3: This protein binds the cAMP response element (CRE) (consensus: 5 -GTGACGT[AC][AG]-3 ), a sequence present in many viral and cellular promoters. Represses transcription from promoters with ATF sites. It may repress transcription by stabilizing the binding of inhibitory cofactors at the promoter. Isoform 2 activates transcription presumably by sequestering inhibitory cofactors away from the promoters. Belongs to the bZIP family. ATF subfamily. 4 isoforms of the human protein are produced by alternative splicing. Protein type: Transcription factor. Chromosomal Location of Human Ortholog: 1q32.3. Cellular Component: nucleoplasm; nucleolus; nucleus. Molecular Function: identical protein binding; protein binding; protein homodimerization activity; protein heterodimerization activity; transcription factor activity; transcription corepressor activity. Biological Process: unfolded protein response, activation of signaling protein activity; cellular protein metabolic process; transcription, DNA-dependent; unfolded protein response; positive regulation of cell proliferation; positive regulation of transcription from RNA polymerase II promoter; negative regulation of transcription from RNA polymerase II promoter; gluconeogenesis
