Principle of the assay: UCP3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-UCP3 antibody and an UCP3-HRP conjugate. The assay sample and buffer are incubated together with UCP3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UCP3 concentration since UCP3 from samples and UCP3-HRP conjugate compete for the anti-UCP3 antibody binding site. Since the number of sites is limited, as more sites are occupied by UCP3 from the sample, fewer sites are left to bind UCP3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UCP3 concentration in each sample is interpolated from this standard curve.

Mitochondrial uncoupling proteins (UCP) are members of the larger family of mitochondrial anion carrier proteins (MACP). UCPs separate oxidative phosphorylation from ATP synthesis with energy dissipated as heat, also referred to as the mitochondrial proton leak. UCPs facilitate the transfer of anions from the inner to the outer mitochondrial membrane and the return transfer of protons from the outer to the inner mitochondrial membrane. They also reduce the mitochondrial membrane potential in mammalian cells. The different UCPs have tissue-specific expression; this gene is primarily expressed in skeletal muscle. This gene's protein product is postulated to protect mitochondria against lipid-induced oxidative stress. Expression levels of this gene increase when fatty acid supplies to mitochondria exceed their oxidation capacity and the protein enables the export of fatty acids from mitochondria. UCPs contain the three solcar protein domains typically found in MACPs. Two splice variants have been found for this gene.[provided by RefSeq, Nov 2008]
