specificity :
This assay has high sensitivity and excellent specificity for detection of TPH1. No significant cross-reactivity or interference between TPH1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TPH1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Sensitivity: 0.1 ng/mL. Assay Type: Competitive
other info2 :
Intended Uses: This TPH1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse TPH1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the Assay: TPH1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-TPH1 antibody and an TPH1-HRP conjugate. The assay sample and buffer are incubated together with TPH1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TPH1 concentration since TPH1 from samples and TPH1-HRP conjugate compete for the anti-TPH1 antibody binding site. Since the number of sites is limited, as more sites are occupied by TPH1 from the sample, fewer sites are left to bind TPH1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TPH1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the aromatic amino acid hydroxylase family. The encoded protein catalyzes the first and rate limiting step in the biosynthesis of serotonin, an important hormone and neurotransmitter. Mutations in this gene have been associated with an elevated risk for a variety of diseases and disorders, including schizophrenia, somatic anxiety, anger-related traits, bipolar disorder, suicidal behavior, addictions, and others.[provided by RefSeq, Apr 2009]
uniprot summary :
TPH1: an enzyme that is rate-limiting in the biosynthesis of serotonin. It catalyzes the biopterin dependent monooxygenation of tryptophan to 5-hydroxytryptophan (5HT), which is subsequently decarboxylated to form the neurotransmitter serotonin. Its expression is limited to a few specialized tissues: raphe neurons, pinealocytes, mast cells, mononuclear leukocytes, beta-cells of the islets of Langerhans, and intestinal and pancreatic enterochromaffin cells. Two alternatively spliced isoforms have been described. Protein type: EC 1.14.16.4; Oxidoreductase; Amino Acid Metabolism - tryptophan. Chromosomal Location of Human Ortholog: 11p15.3-p14. Cellular Component: neuron projection; cytosol. Molecular Function: amino acid binding; tryptophan 5-monooxygenase activity; iron ion binding. Biological Process: circadian rhythm; aromatic amino acid family metabolic process; indolalkylamine biosynthetic process; negative regulation of ossification; positive regulation of fat cell differentiation; bone remodeling; serotonin biosynthetic process
