specificity :
This assay has high sensitivity and excellent specificity for detection of CTNND2. No significant cross-reactivity or interference between CTNND2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CTNND2 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This CTNND2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CTNND2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: CTNND2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CTNND2 antibody and an CTNND2-HRP conjugate. The assay sample and buffer are incubated together with CTNND2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CTNND2 concentration since CTNND2 from samples and CTNND2-HRP conjugate compete for the anti-CTNND2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CTNND2 from the sample, fewer sites are left to bind CTNND2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CTNND2 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene is the tropomyosin-binding subunit of the troponin complex, which is located on the thin filament of striated muscles and regulates muscle contraction in response to alterations in intracellular calcium ion concentration. Mutations in this gene have been associated with familial hypertrophic cardiomyopathy as well as with dilated cardiomyopathy. Transcripts for this gene undergo alternative splicing that results in many tissue-specific isoforms, however, the full-length nature of some of these variants has not yet been determined. uniprot summary :
TNNT2: Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity. Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart. Belongs to the troponin T family. 11 isoforms of the human protein are produced by alternative splicing. Protein type: Motility/polarity/chemotaxis; Motor. Chromosomal Location of Human Ortholog: 1q32. Cellular Component: sarcomere; troponin complex; cytosol; striated muscle thin filament. Molecular Function: troponin C binding; structural constituent of cytoskeleton; troponin I binding; ATPase activity; actin binding; tropomyosin binding. Biological Process: regulation of muscle contraction; metabolic process; positive regulation of ATPase activity; atrial cardiac muscle morphogenesis; ventricular cardiac muscle morphogenesis; sarcomere organization; response to calcium ion; negative regulation of ATPase activity; regulation of heart contraction; muscle filament sliding.
