Porcine
specificity :
This assay has high sensitivity and excellent specificity for detection of AMELX. No significant cross-reactivity or interference between AMELX and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between AMELX and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This AMELX ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine AMELX. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay AMELX ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-AMELX antibody and an AMELX-HRP conjugate. The assay sample and buffer are incubated together with AMELX-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AMELX concentration since AMELX from samples and AMELX-HRP conjugate compete for the anti-AMELX antibody binding site. Since the number of sites is limited, as more sites are occupied by AMELX from the sample, fewer sites are left to bind AMELX-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AMELX concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the amelogenin family of extracellular matrix proteins. Amelogenins are involved in biomineralization during tooth enamel development. Mutations in this gene cause X-linked amelogenesis imperfecta. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]
uniprot summary :
AMELX: Plays a role in biomineralization. Seems to regulate the formation of crystallites during the secretory stage of tooth enamel development. Thought to play a major role in the structural organization and mineralization of developing enamel. Defects in AMELX are the cause of amelogenesis imperfecta type 1E (AI1E). A X-linked defect of dental enamel formation. Teeth have only a thin layer of enamel with normal hardness. The thinness of the enamel makes the teeth appear small. Belongs to the amelogenin family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted, signal peptide; Secreted. Chromosomal Location of Human Ortholog: Xp22.31-p22.1. Cellular Component: proteinaceous extracellular matrix; cell surface. Molecular Function: identical protein binding; protein binding; growth factor activity; hydroxyapatite binding; structural constituent of tooth enamel. Biological Process: osteoblast differentiation; cell proliferation; ion homeostasis; biomineral formation; positive regulation of collagen biosynthetic process; chondrocyte differentiation; epithelial to mesenchymal transition; cell adhesion; signal transduction; odontogenesis of dentine-containing teeth. Disease: Amelogenesis Imperfecta, Type Ie
