specificity :
This assay has high sensitivity and excellent specificity for detection of ACOACA1. No significant cross-reactivity or interference between ACOACA1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ACOACA1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.

Intended Uses: This ACOACA1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ACOACA1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the assay: ACOACA1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACOACA1 antibody and an ACOACA1-HRP conjugate. The assay sample and buffer are incubated together with ACOACA1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACOACA1 concentration since ACOACA1 from samples and ACOACA1-HRP conjugate compete for the anti-ACOACA1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACOACA1 from the sample, fewer sites are left to bind ACOACA1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACOACA1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. Multiple alternatively spliced transcript variants divergent in the 5' sequence and encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
ACC1: a subunit of acetyl-CoA carboxylase (ACC), a multifunctional enzyme system. Catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. Phosphorylation by AMPK or PKA inhibits the enzymatic activity of ACC. ACC-alpha is the predominant isoform in liver, adipocyte and mammary gland. ACC-beta is the major isoform in skeletal muscle and heart. Phosphorylation regulates its activity. Protein type: EC 6.4.1.2; EC 6.3.4.14; Ligase; Carbohydrate Metabolism - pyruvate; Carbohydrate Metabolism - propanoate; Lipid Metabolism - fatty acid biosynthesis. Chromosomal Location of Human Ortholog: 17q21. Cellular Component: mitochondrion; cytoplasm; nucleolus; cytosol; actin cytoskeleton.
