specificity :
This assay has high sensitivity and excellent specificity for detection of SYT-1. No significant cross-reactivity or interference between SYT-1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SYT-1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
other info2 :
Intended Uses: This SYT-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat SYT-1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!
products description :
Principle of the assay: SYT-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SYT-1 antibody and an SYT-1-HRP conjugate. The assay sample and buffer are incubated together with SYT-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SYT-1 concentration since SYT-1 from samples and SYT-1-HRP conjugate compete for the anti-SYT-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SYT-1 from the sample, fewer sites are left to bind SYT-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SYT-1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The synaptotagmins are integral membrane proteins of synaptic vesicles thought to serve as Ca(2+) sensors in the process of vesicular trafficking and exocytosis. Calcium binding to synaptotagmin-1 participates in triggering neurotransmitter release at the synapse (Fernandez-Chacon et al., 2001 [PubMed 11242035]).[supplied by OMIM, Jul 2010]
uniprot summary :
SYT1: an integral membrane protein of synaptic vesicles thought to serve as a Ca(2+) sensor in the process of vesicular trafficking and exocytosis. Calcium binding to synaptotagmin I participates in triggering neurotransmitter release at the synapse. Binds acidic phospholipids with a specificity that requires the presence of both an acidic head group and a diacyl backbone. A Ca(2+)-dependent interaction between synaptotagmin and putative receptors for activated protein kinase C has also been reported. It can bind to at least three additional proteins in a Ca(2+)-independent manner; these are neurexins, syntaxin and AP2. Protein type: Vesicle; Membrane protein, integral; Calcium-binding. Chromosomal Location of Human Ortholog: 12cen-q21. Cellular Component: SNARE complex; presynaptic membrane; synaptic vesicle; neuron projection; synaptic vesicle membrane; integral to membrane; plasma membrane; excitatory synapse; cell junction. Molecular Function: protein C-terminus binding; identical protein binding; clathrin binding; calcium-dependent phospholipid binding; transporter activity; calcium ion binding; calcium-dependent protein binding; calmodulin binding; phosphatidylinositol-4,5-bisphosphate binding; SNARE binding; protein binding; low-density lipoprotein receptor binding; phosphatidylserine binding; syntaxin-1 binding; phosphatidylinositol binding. Biological Process: regulation of exocytosis; positive regulation of calcium ion-dependent exocytosis; synaptic transmission; synaptic vesicle exocytosis; regulation of synaptic transmission, glutamatergic; vesicle docking; glutamate secretion; neurotransmitter secretion; positive regulation of vesicle fusion; pathogenesis; detection of calcium ion; protein homooligomerization; positive regulation of synaptic transmission
