products description :
Principle of the Assay: ACP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACP antibody and an ACP-HRP conjugate. The assay sample and buffer are incubated together with ACP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACP concentration since ACP from samples and ACP-HRP conjugate compete for the anti-ACP antibody binding site. Since the number of sites is limited, as more sites are occupied by ACP from the sample, fewer sites are left to bind ACP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACP concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes an enzyme that catalyzes the conversion of orthophosphoric monoester to alcohol and orthophosphate. It is synthesized under androgen regulation and is secreted by the epithelial cells of the prostate gland. An alternatively spliced transcript variant encoding a longer isoform has been found for this gene. This isoform contains a transmembrane domain and is localized in the plasma membrane-endosomal-lysosomal pathway. [provided by RefSeq, Sep 2008]
uniprot summary :
ACPP: A non-specific tyrosine phosphatase that dephosphorylates a diverse number of substrates under acidic conditions (pH 4-6) including alkyl, aryl, and acyl orthophosphate monoesters and phosphorylated proteins. Has lipid phosphatase activity and inactivates lysophosphatidic acid in seminal plasma. Belongs to the histidine acid phosphatase family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: EC 3.1.3.2; Cofactor and Vitamin Metabolism - riboflavin; Phosphatase; EC 3.1.3.5; Motility/polarity/chemotaxis. Chromosomal Location of Human Ortholog: 3q22.1. Cellular Component: multivesicular body; extracellular space; Golgi cisterna; apical part of cell; lysosomal membrane; plasma membrane; integral to membrane; intracellular; vesicle membrane; nucleus; filopodium; secretory granule. Molecular Function: 5 -nucleotidase activity; choline binding; identical protein binding; acid phosphatase activity; protein homodimerization activity; phosphoric monoester hydrolase activity; thiamin phosphate phosphatase activity. Biological Process: thiamin metabolic process; dephosphorylation; positive regulation of adenosine receptor signaling pathway; nucleotide metabolic process; regulation of sensory perception of pain; adenosine metabolic process; purine base metabolic process
