specificity :
This assay has high sensitivity and excellent specificity for detection of RYR1-Ab. No significant cross-reactivity or interference between RYR1-Ab and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between RYR1-Ab and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This RYR1-Ab ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human RYR1-Ab. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: RYR1-Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing Ryanodine Receptor 1 antigen and an RYR1-Ab-HRP conjugate. The assay sample and buffer are incubated together with RYR1-Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RYR1-Ab concentration since RYR1-Ab from samples and RYR1-Ab-HRP conjugate compete for the Ryanodine Receptor 1 antigen binding site. Since the number of sites is limited, as more sites are occupied by RYR1-Ab from the sample, fewer sites are left to bind RYR1-Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RYR1-Ab concentration in each sample is interpolated from this standard curve.

uniprot summary :
S100A9: a calcium-binding regulatory protein of the S-100 family expressed by macrophages in acutely inflammated tissues and in chronic inflammations. May be an inhibitor of protein kinases. Also expressed in epithelial cells constitutively or induced during dermatoses. May interact with components of the intermediate filaments in monocytes and epithelial cells. Interacts with CEACAM3 in a calcium-dependent manner. Protein type: Calcium-binding. Chromosomal Location of Human Ortholog: 1q21. Cellular Component: extracellular space; cytoskeleton; extracellular region; plasma membrane; nucleus; cytosol. Molecular Function: arachidonic acid binding; antioxidant activity; protein binding; signal transducer activity; RAGE receptor binding; zinc ion binding; microtubule binding; calcium ion binding.
