products description :
Principle of the Assay: ACTHR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ACTHR antibody and an ACTHR-HRP conjugate. The assay sample and buffer are incubated together with ACTHR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTHR concentration since ACTHR from samples and ACTHR-HRP conjugate compete for the anti-ACTHR antibody binding site. Since the number of sites is limited, as more sites are occupied by ACTHR from the sample, fewer sites are left to bind ACTHR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACTHR concentration in each sample is interpolated from this standard curve.

ncbi summary :
MC2R encodes one member of the five-member G-protein associated melanocortin receptor family. Melanocortins (melanocyte-stimulating hormones and adrenocorticotropic hormone) are peptides derived from pro-opiomelanocortin (POMC). MC2R is selectively activated by adrenocorticotropic hormone, whereas the other four melanocortin receptors recognize a variety of melanocortin ligands. Mutations in MC2R can result in familial glucocorticoid deficiency. Alternate transcript variants have been found for this gene. [provided by RefSeq, May 2014]
uniprot summary :
MC2R: Receptor for ACTH. This receptor is mediated by G proteins (G(s)) which activate adenylate cyclase. Defects in MC2R are the cause of glucocorticoid deficiency type 1 (GCCD1); also known as familial glucocorticoid deficiency type 1 (FGD1). GCCD1 is an autosomal recessive disorder due to congenital insensitivity or resistance to adrenocorticotropin (ACTH). It is characterized by progressive primary adrenal insufficiency, without mineralocorticoid deficiency. Belongs to the G-protein coupled receptor 1 family. Protein type: Membrane protein, integral; GPCR, family 1; Receptor, GPCR; Membrane protein, multi-pass. Chromosomal Location of Human Ortholog: 18p11.2. Cellular Component: integral to plasma membrane; cytoplasm; plasma membrane. Molecular Function: protein binding; melanocortin receptor activity; adrenocorticotropin receptor activity. Biological Process: G-protein coupled receptor protein signaling pathway; G-protein signaling, coupled to cyclic nucleotide second messenger; neuropeptide signaling pathway; positive regulation of cAMP biosynthetic process; placenta development. Disease: Glucocorticoid Deficiency 1
