specificity :
This assay has high sensitivity and excellent specificity for detection of CD4+. No significant cross-reactivity or interference between CD4+ and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CD4+ and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.

products description :
Principle of the assay: CD4+ ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CD4+ antibody and an CD4+-HRP conjugate. The assay sample and buffer are incubated together with CD4+-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD4+ concentration since CD4+ from samples and CD4+-HRP conjugate compete for the anti-CD4+ antibody binding site. Since the number of sites is limited, as more sites are occupied by CD4+ from the sample, fewer sites are left to bind CD4+-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD4+ concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigenes and is also a receptor for the human immunodeficiency virus. This gene is expressed not only in T lymphocytes, but also in B cells, macrophages, and granulocytes. It is also expressed in specific regions of the brain. The protein functions to initiate or augment the early phase of T-cell activation, and may function as an important mediator of indirect neuronal damage in infectious and immune-mediated diseases of the central nervous system. Multiple alternatively spliced transcript variants encoding different isoforms have been identified in this gene. [provided by RefSeq]
uniprot summary :
CD4: Accessory protein for MHC class-II antigen/T-cell receptor interaction. May regulate T-cell activation. Induces the aggregation of lipid rafts. Associates with LCK. Binds to HIV-1 gp120 and to P4HB/PDI and upon HIV-1 binding to the cell membrane, is part of P4HB/PDI- CD4-CXCR4-gp120 complex. Interacts with HIV-1 Envelope polyprotein gp160 and protein Vpu. Interacts with Human Herpes virus 7 capsid proteins. Interacts with PTK2/FAK1; this interaction requires the presence of HIV-1 gp120. Protein type: Cell surface; Membrane protein, integral. Chromosomal Location of Human Ortholog: 12p13.31. Cellular Component: T cell receptor complex; endoplasmic reticulum membrane; endoplasmic reticulum lumen; early endosome; integral to membrane; plasma membrane; external side of plasma membrane; lipid raft.
