specificity :
This assay has high sensitivity and excellent specificity for detection of RSPO1. No significant cross-reactivity or interference between RSPO1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between RSPO1 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, cell culture supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 pg/mL.
products description :
Intended Uses: This RSPO1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse RSPO1. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay RSPO1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RSPO1 antibody and an RSPO1-HRP conjugate. The assay sample and buffer are incubated together with RSPO1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RSPO1 concentration since RSPO1 from samples and RSPO1-HRP conjugate compete for the anti-RSPO1 antibody binding site. Since the number of sites is limited, as more sites are occupied by RSPO1 from the sample, fewer sites are left to bind RSPO1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RSPO1 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2014]
uniprot summary :
R-spondin-1: Activator of the beta-catenin signaling cascade, leading to TCF-dependent gene activation. Acts both in the canonical Wnt/beta-catenin-dependent pathway and in non-canonical Wnt signaling pathway, probably by acting as an inhibitor of ZNRF3, an important regulator of the Wnt signaling pathway. Acts as a ligand for frizzled FZD8 and LRP6. May negatively regulate the TGF-beta pathway. Has a essential roles in ovary determination. Defects in RSPO1 are the cause of palmoplantar keratoderma with squamous cell carcinoma of skin and sex reversal (PKKSCC). This recessive syndrome is characterized by XX (female to male) SRY-independent sex reversal, palmoplantar hyperkeratosis and predisposition to squamous cell carcinoma of the skin. Belongs to the R-spondin family. 3 isoforms of the human protein are produced by alternative splicing. Protein type: Secreted; Cell development/differentiation; Cell adhesion; Secreted, signal peptide. Chromosomal Location of Human Ortholog: 1p34.3. Cellular Component: extracellular space; extracellular region; nucleus. Molecular Function: heparin binding; protein binding; G-protein-coupled receptor binding; receptor binding. Biological Process: regulation of gene expression; regulation of receptor internalization; positive regulation of protein amino acid phosphorylation; Wnt receptor signaling pathway through beta-catenin; male meiosis; positive regulation of Wnt receptor signaling pathway. Disease: Palmoplantar Hyperkeratosis With Squamous Cell Carcinoma Of Skin And 46,xx Sex Reversal
