Principle of the assay: GAD67-Ab ELISA kit applies the competitive enzyme immunoassay technique utilizing GAD67 antigen and an GAD67-Ab-HRP conjugate. The assay sample and buffer are incubated together with GAD67-Ab-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAD67-Ab concentration since GAD67-Ab from samples and GAD67-Ab-HRP conjugate compete for the GAD67 antigen binding site. Since the number of sites is limited, as more sites are occupied by GAD67-Ab from the sample, fewer sites are left to bind GAD67-Ab-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAD67-Ab concentration in each sample is interpolated from this standard curve.

This gene encodes one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantigen and an autoreactive T cell target in insulin-dependent diabetes. This gene may also play a role in the stiff man syndrome. Deficiency in this enzyme has been shown to lead to pyridoxine dependency with seizures. Alternative splicing of this gene results in two products, the predominant 67-kD form and a less-frequent 25-kD form. [provided by RefSeq, Jul 2008]
