specificity :
This assay has high sensitivity and excellent specificity for detection of ACE2. No significant cross-reactivity or interference between ACE2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ACE2 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This ACE2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ACE2. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: ACE2 ELISA kit applies the quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with a monoclonal antibody specific for ACE2. Standards or samples are then added to the microtiter plate wells and ACE2 if present, will bind to the antibody pre-coated wells. In order to quantitatively determine the amount of ACE2 present in the sample, a standardized preparation of horseradish peroxidase (HRP)-conjugated polyclonal antibody, specific for ACE2 are added to each well to "sandwich" the ACE2 immobilized on the plate. The microtiter plate undergoes incubation, and then the wells are thoroughly washed to remove all unbound components. Next, substrate solutions are added to each well. The enzyme (HRP) and substrate are allowed to react over a short incubation period. Only those wells that contain ACE2 and enzyme-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACE2 concentration in each sample is interpolated from this standard curve.

ncbi summary :
The protein encoded by this gene belongs to the angiotensin-converting enzyme family of dipeptidyl carboxydipeptidases and has considerable homology to human angiotensin 1 converting enzyme. This secreted protein catalyzes the cleavage of angiotensin I into angiotensin 1-9, and angiotensin II into the vasodilator angiotensin 1-7. The organ- and cell-specific expression of this gene suggests that it may play a role in the regulation of cardiovascular and renal function, as well as fertility. In addition, the encoded protein is a functional receptor for the spike glycoprotein of the human coronaviruses SARS and HCoV-NL63. [provided by RefSeq, Jul 2008]
uniprot summary :
ACE2: Carboxypeptidase which converts angiotensin I to angiotensin 1-9, a peptide of unknown function, and angiotensin II to angiotensin 1-7, a vasodilator. Also able to hydrolyze apelin- 13 and dynorphin-13 with high efficiency. May be an important regulator of heart function. In case of human coronaviruses SARS and HCoV-NL63 infections, serve as functional receptor for the spike glycoprotein of both coronaviruses. Interacts with ITGB1. Interacts with SARS-CoV and HCoV- NL63 spike glycoprotein. Up-regulated in failing heart. Expressed in endothelial cells from small and large arteries, and in arterial smooth muscle cells. Expressed in lung alveolar epithelial cells, enterocytes of the small intestine, Leydig cells and Sertoli cells. Expressed in heart, kidney, testis, and gastrointestinal system. Activated by chloride and fluoride, but not bromide. Inhibited by MLN-4760, cFP_Leu, and EDTA, but not by the ACE inhibitors linosipril, captopril and enalaprilat. Belongs to the peptidase M2 family. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Protease; Membrane protein, integral; EC 3.4.17.23. Chromosomal Location of Human Ortholog: Xp22. Cellular Component: extracellular space; cell surface; cytoplasm; integral to membrane; plasma membrane; extracellular region; lipid raft. Molecular Function: peptidyl-dipeptidase activity; viral receptor activity; protein binding; carboxypeptidase activity; zinc ion binding; metallopeptidase activity; peptide hormone binding; endopeptidase activity; glycoprotein binding. Biological Process: virion attachment, binding of host cell surface receptor; entry of virus into host cell; maternal process involved in pregnancy; regulation of vasodilation; proteolysis; angiotensin maturation; regulation of systemic arterial blood pressure by renin-angiotensin; regulation of cell proliferation; regulation of cytokine production; cellular protein metabolic process; regulation of vasoconstriction; angiotensin catabolic process in blood; regulation of inflammatory response; receptor biosynthetic process; angiotensin mediated drinking behavior
