Principle of the assay: ZYX ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ZYX antibody and an ZYX-HRP conjugate. The assay sample and buffer are incubated together with ZYX-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ZYX concentration since ZYX from samples and ZYX-HRP conjugate compete for the anti-ZYX antibody binding site. Since the number of sites is limited, as more sites are occupied by ZYX from the sample, fewer sites are left to bind ZYX-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ZYX concentration in each sample is interpolated from this standard curve.

Zyxin is a zinc-binding phosphoprotein that concentrates at focal adhesions and along the actin cytoskeleton. Zyxin has an N-terminal proline-rich domain and three LIM domains in its C-terminal half. The proline-rich domain may interact with SH3 domains of proteins involved in signal transduction pathways while the LIM domains are likely involved in protein-protein binding. Zyxin may function as a messenger in the signal transduction pathway that mediates adhesion-stimulated changes in gene expression and may modulate the cytoskeletal organization of actin bundles.
