specificity :
This assay has high sensitivity and excellent specificity for detection of POSTN. No significant cross-reactivity or interference between POSTN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between POSTN and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.

products description :
Intended Uses: This POSTN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine POSTN. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: POSTN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-POSTN antibody and an POSTN-HRP conjugate. The assay sample and buffer are incubated together with POSTN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the POSTN concentration since POSTN from samples and POSTN-HRP conjugate compete for the anti-POSTN antibody binding site. Since the number of sites is limited, as more sites are occupied by POSTN from the sample, fewer sites are left to bind POSTN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The POSTN concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a member of the POU family of transcription factors that regulate mammalian development. The protein regulates expression of several genes involved in pituitary development and hormone expression. Mutations in this genes result in combined pituitary hormone deficiency. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
uniprot summary :
GHF-1: Transcription factor involved in the specification of the lactotrope, somatotrope, and thyrotrope phenotypes in the developing anterior pituitary. Activates growth hormone and prolactin genes. Specifically binds to the consensus sequence 5 - TAAAT-3 . Belongs to the POU transcription factor family. Class- 1 subfamily. 2 isoforms of the human protein are produced by alternative splicing. Protein type: Transcription factor; DNA-binding. Chromosomal Location of Human Ortholog: 3p11. Cellular Component: nucleoplasm; cytoplasm. Molecular Function: chromatin binding; transcription factor activity; transcription corepressor activity. Biological Process: transcription from RNA polymerase II promoter; nuclear transport; regulation of insulin-like growth factor receptor signaling pathway; positive regulation of transcription, DNA-dependent; cell fate specification; positive regulation of inositol trisphosphate biosynthetic process; positive regulation of multicellular organism growth; negative regulation of transcription from RNA polymerase II promoter; determination of adult life span; negative regulation of cell proliferation; regulation of transcription, DNA-dependent; B cell differentiation; positive regulation of cell proliferation; somatotropin secreting cell development; positive regulation of transcription from RNA polymerase II promoter.
