Principle of the assay: ATF-6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ATF-6 antibody and an ATF-6-HRP conjugate. The assay sample and buffer are incubated together with ATF-6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATF-6 concentration since ATF-6 from samples and ATF-6-HRP conjugate compete for the anti-ATF-6 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATF-6 from the sample, fewer sites are left to bind ATF-6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATF-6 concentration in each sample is interpolated from this standard curve.

This gene encodes a transcription factor that activates target genes for the unfolded protein response (UPR) during endoplasmic reticulum (ER) stress. Although it is a transcription factor, this protein is unusual in that it is synthesized as a transmembrane protein that is embedded in the ER. It functions as an ER stress sensor/transducer, and following ER stress-induced proteolysis, it functions as a nuclear transcription factor via a cis-acting ER stress response element (ERSE) that is present in the promoters of genes encoding ER chaperones. This protein has been identified as a survival factor for quiescent but not proliferative squamous carcinoma cells. There have been conflicting reports about the association of polymorphisms in this gene with diabetes in different populations, but another polymorphism has been associated with increased plasma cholesterol levels.
