specificity :
This assay has high sensitivity and excellent specificity for detection of SPRR1B. No significant cross-reactivity or interference between SPRR1B and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between SPRR1B and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 0.1 ng/mL.
other info2 :
Intended Uses: This SPRR1B ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human SPRR1B. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: SPRR1B ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SPRR1B antibody and an SPRR1B-HRP conjugate. The assay sample and buffer are incubated together with SPRR1B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SPRR1B concentration since SPRR1B from samples and SPRR1B-HRP conjugate compete for the anti-SPRR1B antibody binding site. Since the number of sites is limited, as more sites are occupied by SPRR1B from the sample, fewer sites are left to bind SPRR1B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SPRR1B concentration in each sample is interpolated from this standard curve.

uniprot summary :
SPRR1B: Cross-linked envelope protein of keratinocytes. It is a keratinocyte protein that first appears in the cell cytosol, but ultimately becomes cross-linked to membrane proteins by transglutaminase. All that results in the formation of an insoluble envelope beneath the plasma membrane. Can function as both amine donor and acceptor in transglutaminase-mediated cross- linkage. Belongs to the cornifin (SPRR) family. Protein type: Adaptor/scaffold. Chromosomal Location of Human Ortholog: 1q21-q22. Cellular Component: cytoplasm; cornified envelope. Molecular Function: protein binding, bridging; structural molecule activity. Biological Process: keratinocyte differentiation; epidermis development; keratinization; peptide cross-linking
