Intended Uses: This CLDN-18 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rabbit CLDN-18. This ELISA kit for research use only, not for therapeutic or diagnostic applications!. Principle of the Assay: CLDN-18 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CLDN-18 antibody and an CLDN-18-HRP conjugate. The assay sample and buffer are incubated together with CLDN-18-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLDN-18 concentration since CLDN-18 from samples and CLDN-18-HRP conjugate compete for the anti-CLDN-18 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLDN-18 from the sample, fewer sites are left to bind CLDN-18-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN-18 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the claudin family. Claudins are integral membrane proteins and components of tight junction strands. Tight junction strands serve as a physical barrier to prevent solutes and water from passing freely through the paracellular space between epithelial or endothelial cell sheets, and also play critical roles in maintaining cell polarity and signal transductions. This gene is upregulated in patients with ulcerative colitis and highly overexpressed in infiltrating ductal adenocarcinomas. PKC/MAPK/AP-1 (protein kinase C/mitogen-activated protein kinase/activator protein-1) dependent pathway regulates the expression of this gene in gastric cells.
