Principle of the assay: ALOX5AP ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ALOX5AP antibody and an ALOX5AP-HRP conjugate. The assay sample and buffer are incubated together with ALOX5AP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ALOX5AP concentration since ALOX5AP from samples and ALOX5AP-HRP conjugate compete for the anti-ALOX5AP antibody binding site. Since the number of sites is limited, as more sites are occupied by ALOX5AP from the sample, fewer sites are left to bind ALOX5AP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ALOX5AP concentration in each sample is interpolated from this standard curve.

This gene encodes a protein which, with 5-lipoxygenase, is required for leukotriene synthesis. Leukotrienes are arachidonic acid metabolites which have been implicated in various types of inflammatory responses, including asthma, arthritis and psoriasis. This protein localizes to the plasma membrane. Inhibitors of its function impede translocation of 5-lipoxygenase from the cytoplasm to the cell membrane and inhibit 5-lipoxygenase activation. A stroke is an acute neurologic event leading to death of neural tissue of the brain and resulting in loss of motor, sensory and/or cognitive function. Ischemic strokes, resulting from vascular occlusion, is considered to be a highly complex disease consisting of a group of heterogeneous disorders with multiple genetic and environmental risk factors. Genetic variations in ALOX5AP may be associated with susceptibility to myocardial infarction.
