specificity :
This assay has high sensitivity and excellent specificity for detection of VPS53. No significant cross-reactivity or interference between VPS53 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between VPS53 and all the analogues, therefore, cross reaction may still exist in some cases.
storage stability :
Store all reagents at 2-8 degree C.
other info1 :
Samples: Serum, plasma, Cell Culture Supernatants, body fluid and tissue homogenate. Assay Type: Competitive. Sensitivity: 1.0 pg/mL.
other info2 :
Intended Uses: This VPS53 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human VPS53. This ELISA kit for research use only, not for therapeutic or diagnostic applications!

products description :
Principle of the assay: VPS53 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-VPS53 antibody and an VPS53-HRP conjugate. The assay sample and buffer are incubated together with VPS53-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VPS53 concentration since VPS53 from samples and VPS53-HRP conjugate compete for the anti-VPS53 antibody binding site. Since the number of sites is limited, as more sites are occupied by VPS53 from the sample, fewer sites are left to bind VPS53-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VPS53 concentration in each sample is interpolated from this standard curve.

ncbi summary :
This gene encodes a protein with sequence similarity to the yeast Vps53p protein. Vps53p is involved in retrograde vesicle trafficking in late Golgi. [provided by RefSeq, Jul 2008]
uniprot summary :
VPS53: May be involved in retrograde transport of early and late endosomes to the late Golgi. The GARP complex is required for the maintenance of the cycling of mannose 6-phosphate receptors between the TGN and endosomes, this cycling is necessary for proper lysosomal sorting of acid hydrolases such as CTSD. Belongs to the VPS53 family. 4 isoforms of the human protein are produced by alternative splicing. Chromosomal Location of Human Ortholog: 17p13.3. Cellular Component: Golgi apparatus; perinuclear region of cytoplasm; endosome membrane. Molecular Function: protein binding. Biological Process: protein transport; retrograde transport, endosome to Golgi. Disease: Pontocerebellar Hypoplasia, Type 2e
