Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for COLVI has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any COLVI present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for COLVI is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of COLVI bound in the initial step. The color development is stopped and the intensity of the color is measured.

Collagens are extracellular matrix proteins and have a triple-helical domain as their common structural element. Collagen VI is a major structural component of microfibrils. The basic structural unit of collagen VI is a heterotrimer of the alpha1(VI), alpha2(VI), and alpha3(VI) chains. The alpha2(VI) and alpha3(VI) chains are encoded by the COL6A2 and COL6A3 genes, respectively. The protein encoded by this gene is the alpha 1 subunit of type VI collagen (alpha1(VI) chain).
